چکیده

Tumor-associated macrophages promote tumor growth by stimulating angiogenesis and suppressing antitumor immunity. Thus, therapeutics that inhibit macrophage recruitment to tumors may provide new avenues for cancer therapy. In this study, we showed how chemoattractants stromal cell-derived growth factor 1 alpha (SDF-1a) and interleukin 1 beta (IL-1b) collaborate with myeloid cell integrin-a4b1 to promote tumor inflammation and growth. We found that SDF-1a and IL-1b are highly expressed in the microenvironments of murine lung, pancreatic, and breast tumors; surprisingly, SDF-1a was expressed only by tumor cells, whereas IL-1b was produced only by tumor-derived granulocytes and macrophages. In vivo, both factors directly recruited proangiogenic macrophages to tissues, whereas antagonists of both factors suppressed tumor inflammation, angiogenesis, and growth. Signals induced by IL-1b and SDF-1a promoted the interaction of talin and paxillin with the cytoplasmic tails of integrin-a4b1, thereby stimulating myeloid cell adhesion to endothelium in vitro and in vivo. Inhibition of integrin-a4b1, SDF-1a, or IL-1b was sufficient to block tumor inflammation and growth, and the combined blockade of these molecules greatly accentuated these effects. Furthermore, antagonists of integrin-a4b1 inhibited chemotherapy-induced tumor inflammation and acted synergistically with chemotherapeutic agents to suppress tumor inflammation and growth. These results show that targeting myeloid cell recruitment mechanisms can be an effective approach to suppress tumor progression. Cancer Res; 71(22): 1–11. 2011 AACR.

HEMATOPOIESIS Interleukin - 1 b and Tumor Necrosis Factor - a Stimulate DNA Binding of Hypoxia - Inducible Factor - 1

The rate of transcription of several genes encoding proteins involved in O2 and energy homeostasis is controlled by hypoxia-inducible factor-1 (HIF-1), a heterodimeric DNA binding complex composed of a and b subunits. HIF-1 is considered the primary trans-acting factor for the erythropoietin (EPO) and vascular endothelial growth factor (VEGF) genes. Since EPO gene expression is inhibited by the...

O 2: Anti-Inflammatory Approach to Epilepsy Treatment

Epilepsy is one of the most common neurologic diseases around the world and more significantly in Iran (0.4-1 % worldwide and 5% in Iran). Almost one-third of these patients suffer from treatment-resistant epilepsy, which reduces their quality of life by recurring epileptic onsets. There are different approaches for the treatment of both treatment-resistant and treatment-nonresistant epilepsy, ...

Evaluation of frequency polymorphism Interleukin 1B gene in patients with peptic ulcer and chronic gastritis

Introduction: Helicobacter pylori is the main cause of chronic inflammation and peptic ulcer. More than half of the world population is afflicted with Helicobacter, while only 20% of people show the clinical symptoms of the disease. Genes encoding the cytokines interleukin (IL)-1B play a key role in the inflammatory responses of gastric mucus. This study aims to analyze polymorphism in the IL-1...
